The Big Picture

  • AstraZeneca and former partner Bristol-Myers Squibb began marketing Onglyza in 2009
    before conducting clinical trials to see whether it increased patients’ cardiac risks.
  • In 2008 the FDA guidance urged companies to “demonstrate that” new therapies “will not
    result in an unacceptable increase in cardiovascular risk.”
  • The FDA is concerned about the sufficiency of the Onglyza clinical trials and ordered
    AstraZeneca to perform a post-marketing study.
  • AstraZeneca completed a 16,000 patient cardiovascular outcomes trial called SAVOR, the
    results of which raised major Onglyza safety concerns.
  • The SAVOR trial found that Onglyza patients were 27% more likely to be hospitalized for heart
    failure.
  • The SAVOR study also found a significant increase in the rate of “all-cause mortality” among
    Onglyza patients.
  • In an April 2016 drug safety communication, the FDA announced that Onglyza and Kombiglyze
    XR are now required to carry warning about heart failure risk.
